The elasticity and jawline numbers come from the supplier's own uncontrolled in-vivo panels. The elastase-inhibition mechanism is plausible; the progerin-reduction claim has no independent confirmation in human skin.
How it works
Progerin is an abnormally processed form of lamin A that destabilises the nuclear envelope; it drives Hutchinson-Gilford progeria and accumulates slowly in normally ageing cells. Trifluoroacetyl tripeptide-2 is presented as a biomimetic of elafin, the endogenous elastase inhibitor, and is claimed to both inhibit elastase and MMP activity and to lower progerin levels in fibroblasts. Preserving elastin fibres is a defensible target for jawline and neck laxity, since elastin is not meaningfully replaced in adult skin once it is degraded. The progerin arm of the story is the weaker one — reducing a nuclear lamin intermediate topically is a large claim on modest supplier data.

Mechanism in depth
Progeline is sold on two claims of very unequal quality, and separating them is the whole job. The elastase claim is the credible one. Skin elasticity depends on elastin fibres that are laid down in childhood and essentially never replaced in adult dermis — elastin has a half-life measured in decades and adult fibroblasts barely transcribe it. Neutrophil elastase and macrophage elastase (MMP-12) degrade those fibres, and photoageing and chronic low-grade inflammation both increase elastase activity. So an elastase inhibitor is targeting the one structural protein you genuinely cannot rebuild, and preservation is the only available strategy. Elafin is the endogenous inhibitor and a biomimetic of it is a reasonable design goal. Whether a trifluoroacetylated tripeptide achieves meaningful elastase inhibition at concentrations reachable in human dermis is unpublished, but the target is sound and it explains why the supplier positions this for jawline and neck laxity rather than for wrinkles. The progerin claim is the weak one. Progerin is an aberrantly farnesylated, incompletely processed form of lamin A that arises from cryptic splice site use in LMNA. It causes Hutchinson-Gilford progeria in its extreme form, and it does genuinely accumulate at low levels in normally ageing cells — Takeuchi and Rünger showed that longwave UV directly induces progerin in human skin, which is a real and interesting result that gives the topical application some rationale. But progerin sits in the nuclear lamina. Reducing it means altering splicing or farnesylation or proteolytic clearance inside the nucleus of a dermal fibroblast, from a peptide applied to the skin surface. That is a very large claim resting on supplier fibroblast data. Treat the elastase arm as plausible and the progerin arm as marketing that happens to sit on top of real biology.
What usually goes wrong
People buy this for the wrong anatomy. Jawline and neck laxity in someone over fifty is largely fat pad descent, ligament laxity and bone resorption, and no topical touches any of that. The elastin fraction of the problem is the only part this could plausibly address, and it is a minority of what you see in the mirror. The second mistake is photographic: front-on photos will not show jawline change, so shoot in fixed profile with a fixed head position or you will never know whether it worked. Third, the progerin story attracts people from the longevity world who over-index on a mechanism that has not been shown to happen in human skin from a topical.

What to expect, and when
Week 4: supplier panels report elasticity changes. Week 8: supplier panels report jawline contour change. Week 12-24: realistic assessment window. Because the mechanism is preservation, a substantial part of any benefit is loss that did not happen, which never shows up in a photograph.
Stacking and comparisons
No chemical incompatibilities. The pairing that makes sense is Progeline with a collagen-building matrikine, because they address genuinely different structural failures: elastin degradation causes laxity and recoil loss, collagen loss causes thinning and creasing, and no single peptide addresses both. Pairing with GHK-Cu is also coherent since GHK-Cu shifts the MMP/TIMP balance from a different direction. Because the mechanism is preservation rather than construction, the most important co-intervention is not another peptide at all — it is daily broad-spectrum sunscreen including UVA protection, since UVA drives both the elastase activity and, per Takeuchi, the progerin induction this product claims to counter.
Against the collagen peptides, Progeline targets a protein you cannot rebuild rather than one you can, which makes it strategically different rather than better or worse — preservation is genuinely valuable for elastin because there is no second chance. Against a retinoid, retinoids have actual histological data on elastic fibre morphology in photoaged skin and Progeline has supplier panels. Against energy-based tightening devices for laxity, no topical competes. Against Hexapeptide-11, both are pitched at cellular ageing and Hexapeptide-11 has the independent academic paper.

Genuinely uncertain
